I've got 6.5" woofers in my doors, it required a tiny bit of grinding to slide it in, maybe 1-2mm around the hole to get it in nicely.
I think it really depends on the actual speaker, some will some won't.

I'm honestly not sure, Mine are Phoenix Gold Tantrums, they nudge the window guide, but don't hit the actual window.
you have to be careful with using spacers too, u can use real thin ones, but then you've gotta ensure the speaker doesn't rest against the door trim.

I had to use spacers because the Jaycar spilts are huge. no major mods other than screwing the spacers on.
good luck

Most E series cars have a mounting clearance of about 68-70mm, any deeper and you will need spacers.
The Response speakers are very deep and will definately need spacers.

if u use spaces, wont the speakers stick out past the door trim?? tho i suppose its ok as long as they are firmly mounted, and have a grille for protection?

My Quarts had no problems sliding into place with only some small file work on the door skin to miss the basket, however i had problems with the cone and suspension hitting the door trim. I had to pack the door trim out from the inner skin as my front drivers had enough throw to touch the trim.
